Candidate factors have been identified by the observation that changes in glucocorticoid induction parameters in CV 1 cells could be reproduced by varying the cellular levels of coactivators [ transcriptional intermediary factor 2 ( TIF 2 ) , steroid receptor coactivator 1 ( SRC 1 ) , and amplified in breast cancer 1 ( AIB 1 ) ] , comodulator [ CREB binding protein ( CBP ) ] , or corepressor [ silencing mediator for retinoid and thyroid hormone receptors ( SMRT ) ] without concomitant increases in GR . ^^^ Similarly , additional SMRT could reverse the action of increased levels of GRs in HeLa cells , thus indicating that the effects of cofactors on transcription may be general for GR in a variety of cells . ^^^

Glucocorticoid receptor ( GR ) associated SMRT binding to C / EBPbeta TAD and Nrf 2 Neh4 / 5 : role of SMRT recruited to GR in GSTA 2 gene repression . ^^^ Glucocorticoid receptor ( GR ) associated SMRT binding to C / EBPbeta TAD and Nrf 2 Neh4 / 5 : role of SMRT recruited to GR in GSTA 2 gene repression . ^^^ Immunoprecipitation immunoblotting , chromatin immunoprecipitation , and GST pull down assays revealed that silencing mediator for retinoid and thyroid hormone receptors ( SMRT ) , a corepressor recruited to steroid GR complex for histone deacetylation , bound to TAD domain of C / EBPbeta and Neh4 / 5 domain of Nrf 2 . ^^^ In conclusion , dexamethasone antagonizes C / EBPbeta and Nrf 2 mediated GSTA 2 gene induction via ligand GR binding to the GRE , and steroid mediated GSTA 2 repression involves inactivation of C / EBPbeta and Nrf 2 by SMRT recruited to steroid GR complex . . ^^^

In coexpression studies , the glucocorticoid receptor interacting protein ( GRIP ) coactivator protein and the silencing mediator of retinoid and thyroid ( SMRT ) corepressor protein form spherical subnuclear focal bodies that are spatially distinct , suggesting that specific protein interactions concentrate these divergent proteins in separate subnuclear regions . ^^^
