| In an effort to determine whether the angiopoietins and Tie receptors play a role in the pathobiology of angiosarcoma and KS , we studied the expression of angiopoietin 1 , angiopoietin 2 , angiopoietin 4 , Tie 1 , and Tie 2 mRNAs in biopsies of KS from 12 AIDS patients , in biopsies of cutaneous angiosarcoma from two patients , and in control biopsies of normal skin from three volunteers by in situ hybridization . ^^^ |

| To determine the significance and regulation of the angiopoietin ( Ang ) pathway in highly vascular human renal cell carcinomas ( RCCs ) , this study has investigated the expression of the Ang 1 , Ang 2 , Ang 4 , and Tie 2 genes in a series of normal ( n = 26 ) and neoplastic ( n = 45 ; clear cell n = 35 , papillary n = 10 ) human kidney tissues , examined the pattern of Ang 2 and Tie 2 protein expression , and correlated expression with clinicopathological variables . ^^^ Ang 1 , Ang 2 and Tie 2 , but not Ang 4 mRNA , were detected in normal and tumour samples . ^^^ |

| The angiopoietin ( Ang ) family of growth factors includes Ang 1 , Ang 2 , Ang 3 , and Ang 4 , all of which bind to the endothelial receptor tyrosine kinase Tie 2 . ^^^ In experiments with human endothelial cell lines , Ang 3 was identified as an antagonist of Tie 2 and Ang 4 was identified as an agonist of Tie 2 . ^^^ Ang 4 ( 400 ng / mL ) markedly increased Tie 2 and Akt phosphorylation in primary cultured HUVECs whereas Ang 3 ( 400 ng / mL ) did not produce significant changes . ^^^ Unexpectedly , intravenously administered Ang 3 ( 30 microg ) was more potent than Ang 4 ( 30 microg ) in phosphorylating the Tie 2 receptor in lung tissue from mice in vivo . ^^^ Thus , Ang 3 and Ang 4 are agonists of Tie 2 , but mouse Ang 3 has strong activity only on endothelial cells of its own species . . ^^^ |
