Here we have investigated the possibility that JIP 1 may also affect the catalytic activity of Akt 1 , a serine / threonine kinase that has been implicated in multiple cellular processes , including survival and proliferation . ^^^ JIP 1 expression enhanced Akt 1 kinase activity in a dose dependent manner following serum starvation in 293 cells . ^^^ Cellular activation of Akt 1 following stimulation with low concentrations of insulin like growth factor ( IGF 1 ) was elevated in the presence of JIP 1 . ^^^ JIP 1 expression also prolonged Akt 1 stimulation after a short IGF 1 pulse . ^^^ The mechanism of JIP 1 mediated Akt 1 activation involved JIP 1 protein binding to the Akt 1 pleckstrin homology domain , which in turn promoted the phosphorylation of the activation T loop of Akt 1 by phosphoinositide dependent kinase 1 . ^^^

In this study , we describe a novel interaction between Akt 1 and JNK interacting protein 1 ( JIP 1 ) . ^^^ We first detected the interaction of Akt 1 and JIP 1 in hippocampus at various time points of ischemia . ^^^ In the basal state , JIP 1 bind to Akt 1 , MLK 3 at maximum while JIP 1 binds to JNK 3 at minimum . ^^^ Ischemia stimulus decreased the Akt 1 JIP 1 interaction and concomitantly increased association between JIP 1 and JNK 3 . ^^^ While MLK 3 binding to JIP 1 decreased , similar to Akt 1 JIP 1 interaction during ischemia . ^^^
