Methyl CpG binding protein MBD 1 couples histone H 3 methylation at lysine 9 by SETDB 1 to DNA replication and chromatin assembly . ^^^ Moreover , during DNA replication , MBD 1 recruits SETDB 1 to the large subunit of chromatin assembly factor CAF 1 to form an S phase specific CAF 1 / MBD1 / SETDB1 complex that facilitates methylation of H 3 K9 during replication coupled chromatin assembly . ^^^ Our data suggest a model in which H 3 K9 methylation by SETDB 1 is dependent on MBD 1 and is heritably maintained through DNA replication to support the formation of stable heterochromatin at methylated DNA . . ^^^ |

The histone methyltransferase SETDB 1 and the DNA methyltransferase DNMT3A interact directly and localize to promoters silenced in cancer cells . ^^^ Here , we show that SETDB 1 associates with endogenous DNA methyltransferase activity . ^^^ SETDB 1 interacts with the de novo DNA methyltransferases DNMT3A and DNMT3B but not with the maintenance methyltransferase DNMT 1 . ^^^ In summary , our data demonstrate the direct physical interaction and functional connection between the H 3 K9 trimethylase SETDB 1 and the DNA methyltransferase DNMT3A and thus contribute to a better understanding of the complexity of the self reinforcing heterochromatin machinery operating at silenced promoters . . ^^^ |
