| DISC 1 ( Disrupted In Schizophrenia 1 ) is a centrosome associated protein that interacts with MAP1A , MIPT 3 , ATF4 / 5 and NUDEL : regulation and loss of interaction with mutation . ^^^ DISC 1 interacts by yeast two hybrid , mammalian two hybrid , and co immunoprecipitation assays with multiple proteins of the centrosome and cytoskeletal system , including MIPT 3 , MAP1A and NUDEL ; proteins which localize receptors to membranes , including alpha actinin 2 and beta 4 spectrin ; and proteins which transduce signals from membrane receptors , including ATF 4 and ATF 5 . ^^^ Truncated mutant DISC 1 fails to interact with ATF 4 , ATF 5 or NUDEL . ^^^ Deletion mapping demonstrated that DISC 1 has distinct interaction domains : MAP1A interacts via its LC 2 domain with the N terminus of DISC 1 , whereas MIPT 3 and NUDEL bind via their C terminal domains to the central coiled coil domain of DISC 1 , and ATF4 / 5 bind via their C terminal domains to the C terminus of DISC 1 . ^^^ |

| Recently , nuclear distribution element like ( NUDEL ) has been implicated to play a role in lissencephaly and schizophrenia through interactions with the lissencephaly gene 1 ( Lis 1 ) and disrupted in schizophrenia 1 ( DISC 1 ) products , respectively . ^^^ Cys 273 is very close to the DISC 1 binding site on NUDEL oligopeptidase . ^^^ Intriguingly , DISC 1 inhibits NUDEL oligopeptidase activity in a competitive fashion . ^^^ We suggest that the activity of NUDEL oligopeptidase is under tight regulation through protein protein interactions and that disruption of these interactions , as postulated in a Scottish DISC 1 translocation schizophrenia cohort , may lead to aberrant regulation of NUDEL oligopeptidase , perhaps providing a substrate for the pathology of schizophrenia . . ^^^ |

| In cell and animal models , C terminus truncated DISC 1 disrupts intracellular transport , neural architecture and migration , perhaps because it fails to interact with binding partners involved in neuronal differentiation such as fasciculation and elongation protein zeta 1 ( FEZ 1 ) , platelet activating factor acetylhydrolase , isoform Ib , PAFAH1B1 or lissencephaly 1 protein ( LIS 1 ) and nuclear distribution element like ( NUDEL ) . ^^^ However , the expression of NUDEL , FEZ 1 and LIS 1 was each significantly reduced in the brain tissue from patients with schizophrenia and expression of each showed association with high risk DISC 1 polymorphisms . ^^^ |
