| We studied the role of PDZ proteins GRIP , ABP , and PICK 1 in GluR 2 AMPA receptor trafficking . ^^^ Notably , a single residue substitution , S880A , which blocks binding to ABP / GRIP but not to PICK 1 , reduced synaptic accumulation to the same extent as the PDZ site truncation . ^^^ We conclude that the association of GluR 2 with ABP and / or GRIP but not PICK 1 is essential for maintaining the synaptic surface accumulation of the receptor , possibly by limiting its endocytotic rate . . ^^^ |

| The PDZ proteins PICK 1 , GRIP , and syntenin bind multiple glutamate receptor subtypes . ^^^ Using sequence homology searches , yeast two hybrid assays and glutathione S transferase ( GST ) pull down approaches we have identified a series of glutamate receptor subunits that interact differentially with the PDZ proteins GRIP , PICK 1 , and syntenin . ^^^ We report several receptor protein interactions , strong ones include : ( 1 ) GRIP and syntenin with mGluR7a , mGluR4a , and mGluR 6 ; ( 2 ) PICK 1 and GRIP with mGluR 3 ; and ( 3 ) syntenin with all forms of GluR 1 4 and mGluR7b . ^^^ The wide range of targets for PICK 1 , GRIP , and syntenin suggests they may represent a molecular mechanism that can concentrate and / or regulate a number of different receptors at a common site on a synapse . ^^^ |

| To determine whether gene expression of AMPARs and / or AMPAR binding proteins , which control response / sensitivity of AMPAR bearing neurons to glutamate , are altered in schizophrenia , mRNA expression and abundance of AMPAR subunits ( GluR 1 4 ) and several AMPAR binding proteins ( SAP 97 , PICK 1 , GRIP , ABP ) were measured in the dorsolateral prefrontal cortex ( DLPFC ) and the occipital cortex of elderly schizophrenia patients ( n = 36 ) and matched normal controls ( n = 26 ) by quantitative real time PCR . ^^^ The data also demonstrated significant differences in the abundances of mRNAs encoding GluR 1 4 subunits ( GluR 2 > GluR 3 > GluR 1 > GluR 4 ) and of AMPAR binding proteins ( SAP 97 > PICK 1 > GRIP > ABP ) in both diagnostic groups . ^^^ |
