| We identified the PDZ domain protein PICK 1 as an interactor of BNaC 1 ( ASIC 2 ) in a yeast two hybrid screen . ^^^ The PDZ domain protein PICK 1 and the sodium channel BNaC 1 interact and localize at mechanosensory terminals of dorsal root ganglion neurons and dendrites of central neurons . ^^^ The interaction between BNaC 1 and PICK 1 requires the PDZ domain of PICK 1 and the last four amino acids of BNaC 1 . ^^^ BNaC 1 is similar to two other BNaC / ASIC family members , BNaC 2 ( ASIC 1 ) and ASIC 4 , at its extreme C terminus , and we show that PICK 1 also interacts with BNaC 2 . ^^^ We found that PICK 1 , like BNaC 1 and BNaC 2 , is expressed by DRG neurons and , like the BNaC1alpha isoform , is present at their peripheral mechanosensory endings . ^^^ |

| Protein kinase C stimulates the acid sensing ion channel ASIC2a via the PDZ domain containing protein PICK 1 . ^^^ Recently , the PDZ domain containing protein PICK 1 ( protein interacting with C kinase ) has been shown to interact with ASIC1a and ASIC2a , raising the possibility that protein kinase C ( PKC ) could regulate ASICs . ^^^ We now show that the amplitude of the ASIC2a current , which was only modestly increased ( approximately +30 % ) by the PKC activator 1 oleyl 2 acetyl sn glycerol ( OAG , 50 microm ) in the absence of PICK 1 , was strongly potentiated ( approximately +300 % ) in the presence of PICK 1 . ^^^ This PICK 1 dependent regulatory effect was inhibited in the presence of a PKC inhibitory peptide and required the PDZ domain of PICK 1 as well as the PDZ binding domain of ASIC2a . ^^^ We have also shown the direct PICK 1 dependent phosphorylation of ASIC2a by [ ( 32 ) P ] phosphate labeling and immunoprecipitation and identified a major phosphorylation site , ( 39 ) TIR , on the N terminus part of ASIC2a . ^^^ |
