Functional dissection of transcription factor ZBRK 1 reveals zinc fingers with dual roles in DNA binding and BRCA 1 dependent transcriptional repression . ^^^ To clarify this issue , we have examined the functional interaction between BRCA 1 and ZBRK 1 , a BRCA 1 dependent KRAB eight zinc finger transcriptional repressor . ^^^ We report here the identification and molecular characterization of a portable BRCA 1 dependent transcriptional repression domain within ZBRK 1 composed of zinc fingers 5 8 along with sequences in the unique ZBRK 1 C terminus . ^^^ This C terminal repression domain functions in a BRCA 1 , histone deacetylase , and promoter specific manner and is thus functionally distinguishable from the N terminal KRAB repression domain in ZBRK 1 , which exhibits no BRCA 1 dependence and broad promoter specificity . ^^^ Significantly , we also find that the BRCA 1 dependent transcriptional repression domain on ZBRK 1 includes elements that modulate its sequence specific DNA binding activity . ^^^

Sequence specific transcriptional corepressor function for BRCA 1 through a novel zinc finger protein , ZBRK 1 . ^^^ To explore the mechanistic basis for this regulation , a novel human gene , ZBRK 1 , which encodes a 60 kDa protein with an N terminal KRAB domain and eight central zinc fingers , was identified by virtue of its interaction with BRCA 1 in vitro and in vivo . ^^^ ZBRK 1 binds to a specific sequence , GGGxxx CAGxxxTTT , within GADD 45 intron 3 that supports the assembly of a nuclear complex minimally containing both ZBRK 1 and BRCA 1 . ^^^ ZBRK 1 represses transcription through this recognition sequence in a BRCA 1 dependent manner . ^^^

Previously , we identified a novel zinc finger protein , ZBRK 1 , which contains a KRAB domain at the N terminus , eight zinc fingers at the center , and a BRCA 1 binding region at the C terminus . ^^^ In a BRCA 1 dependent manner , ZBRK 1 represses Gadd45a transcription through binding to a specific sequence in intron 3 . ^^^ In both BRCA 1 proficient and deficient cells , ZBRK 1 is degraded with similar efficiencies independent of BRCA 1 E3 ligase activity . ^^^

The GADD 45 , ZBRK 1 and BRCA 1 pathway : quantitative analysis of mRNA expression in colon carcinomas . ^^^ This gene is a target for coordinate regulation by both ZBRK 1 and BRCA 1 . ^^^ In this study , the relationships between GADD 45 , ZBRK 1 , and BRCA 1 expression were investigated in colon carcinomas . mRNA expression of these three genes was analysed in 116 colon carcinomas by real time reverse transcriptase polymerase chain reaction ( RT PCR ) . ^^^ Possible relationships between expression levels of GADD 45 , ZBRK 1 , and BRCA 1 , and a series of clinicopathological parameters classically associated with poor prognosis , were also examined . ^^^ ZBRK 1 showed a tendency towards underexpression , while GADD 45 and BRCA 1 were generally overexpressed . ^^^
