All three proteins exert their inhibitory activity by binding to a neuronal receptor complex containing the Nogo 66 receptor ( NgR ) and the neurotrophin ( NT ) receptor p75NTR . ^^^ In their recent publication , Mi et al . identify the novel protein Lingo 1 as an interactor of p75NTR and NgR . ^^^ The Lingo 1 NgR p75NTR complex is shown to confer the inhibitory effects on nerve cell regeneration of Nogo 66 , OMgP , and MAG by activating the small guanosine triphosphatase ( GTPase ) RhoA . ^^^

The central nervous system myelin components oligodendrocyte myelin glycoprotein , myelin associated glycoprotein and the Nogo 66 domain of Nogo A inhibit neurite outgrowth by binding the neuronal glycosyl phosphatidylinositol anchored Nogo 66 receptor ( NgR ) that transduces the inhibitory signal to the cell interior via a transmembrane co receptor , p75NTR . ^^^ The N terminal fragment bound Nogo 66 and blocked Nogo 66 binding to cell surface NgR but failed to associate with p75NTR , indicative of a role as a Nogo 66 antagonist . ^^^

Disinhibition of neurotrophin induced dorsal root ganglion cell neurite outgrowth on CNS myelin by siRNA mediated knockdown of NgR , p75NTR and Rho A . ^^^ Using RNA interference ( RNAi ) in dorsal root ganglia neurons ( DRGN ) , we demonstrate siRNA mediated silencing of components of the inhibitory signalling cascade , including p75NTR , NgR and Rho A mRNA , of 70 % , 100 % and 100 % of the relevant protein , respectively , while changes in neither protein levels nor cellular immunoreactivity were detected using the relevant scrambled siRNA control sequences . ^^^ Importantly , after 48 h in culture after siRNA mediated knockdown of Rho A , neurite outgrowth was enhanced by 30 % compared to that after p75NTR and 50 % after NgR silencing . ^^^ By 3 days , a 5 , 3 . 5 and 6 . 5 fold increase in betaIII tubulin protein levels were observed compared to controls without siRNA after knockdown of p75NTR , NgR and Rho A , respectively . ^^^

Schwann cell derived factor induced modulation of the NgR / p75NTR / EGFR axis disinhibits axon growth through CNS myelin in vivo and in vitro . ^^^ When associated with the Nogo receptor ( NgR ) , the transmembrane receptor p75NTR signals growth cone collapse . ^^^ Here , we show that optic nerve regeneration in vivo correlates with Schwann cell derived factor induced cleavage of NgR and Nogo A , and inactivation of p75NTR signalling by the induction of regulated intramembranous proteolysis ( RIP ) and the release of both extracellular ( p75ECD ) and intracellular ( p75ICD ) domains . ^^^ Hence , Schwann cell derived factors compromise inhibitory signalling by ( 1 ) antagonizing ligand / NgR binding with metalloproteinase cleaved Nogo A peptides ; ( 2 ) RIP of p75NTR ; ( 3 ) competitively blocking NgR / p75NTR clustering with soluble p75ECD ; and ( 4 ) consequent reduced downstream EGFR phosphorylation and suppression of Rho A activation . ^^^ Moreover , in RGC cultures , exogenous tumour necrosis converting enzyme ( TACE ) initiates RIP of p75NTR , reduces EGFR phosphorylation , suppresses activation of Rho A , cleaves the ECD from both NgR and TROY , and disinhibits neurotrophic factor ( NTF ) stimulated RGC neurite outgrowth in the presence of CNS myelin . ^^^
