Previous studies have indicated that Smad 5 , together with Smad 1 and Smad 8 , participates in signaling downstream of BMP receptors . ^^^ We further used the Smad 7 promoter sequence that contains a Smad binding element ( SBE ) , GTCTAGAC to determine how mutations in each nucleotide in the SBE affects the binding with Smad 5 , compared with the binding with Smad 1 , Smad 2 , Smad 3 , Smad 4 , and Smad 8 . ^^^ Interestingly , Smad 5 , but not Smad 1 and Smad 8 , was able to bind the SBE , at a level similar to the binding by Smad 3 and Smad 4 . ^^^ These studies suggest that even though Smad 5 falls into the same subfamily with Smad 1 and Smad 8 in mediating the signaling by BMP receptors , it has an unique DNA binding property that is similar to Smad 3 , which specifically transduces signaling for TGF beta and activin receptors . . ^^^

Core binding factor alpha 1 ( Cbfa 1 ) induces osteoblastic differentiation of C2C12 cells without interactions with Smad 1 and Smad 5 . ^^^ In this study we examine the effects of Cbfa 1 on osteoblastic differentiation in the presence or absence of interactions with Smad 1 or Smad 5 using C2C12 undifferentiated mesenchymal cells . ^^^ Introduction of Cbfa 1 into C2C12 cells induced osteoblastic differentiation and promoted transactivation of osteocalcin gene promoter without forming the complex with Smad 1 or Smad 5 . ^^^ These results suggest that the interactions with Smad 1 or Smad 5 are not essential for Cbfa 1 to demonstrate its osteogenic actions . ^^^
