| Guided by Hop structural data and sequence alignment analyses , we have used site directed mutagenesis , co precipitation assays , circular dichroism spectroscopy , steady state fluorescence , and surface plasmon resonance spectroscopy to both qualitatively and quantitatively characterize the contacts necessary for the N terminal tetratricopeptide repeat domain ( TPR 1 ) of mSTI 1 to bind to heat shock cognate protein 70 ( Hsc 70 ) and to discriminate between Hsc 70 and Hsp 90 . ^^^ These results suggest that mSTI 1 Hsc70 interaction requires a network of interactions not only between charged residues in the TPR 1 domain of mSTI 1 and the EEVD motif of Hsc 70 but also outside the TPR domain . ^^^ Using a truncated derivative of mSTI 1 incapable of binding to Hsp 90 , we substituted residues on TPR 1 potentially involved in hydrophobic contacts with Hsc 70 . ^^^ Our results suggest that binding of Hsc 70 to TPR 1 is more specific than binding of Hsp 90 to TPR2A with serious implications for the mechanisms of mSTI 1 interactions with Hsc 70 and Hsp 90 in vivo . . ^^^ |

| Replacement of a bulky hydrophobic residue in TPR 1 disrupted the interaction of mSTI 1 with hsc 70 . ^^^ These results are consistent with a model in which conserved residues within the N terminal TPR region of mSTI 1 contribute differentially to the interaction with hsc 70 , and in which TPR 1 has a significant role in targeting mSTI 1 to hsc 70 . ^^^ |

| In the present study , the effect of the TPR containing proteins , including the C terminus of hsc 70 interacting protein ( CHIP ) , TPR 1 and human glutamine rich TPR containing protein ( hSGT ) , on refolding of luciferase by DnaJ and hsc 70 was investigated . ^^^ The inhibitory effect exerted by TPR 1 and hSGT depended upon their binding to hsc 70 . ^^^ We also demonstrate that the peptide , GPTIEEVD , corresponding to the C terminal end of hsc 70 , abolished the association of [ ( 3 ) H ] hsc 70 with CHIP , TPR 1 and hSGT . ^^^ |
