Here , we demonstrate that three baculovirus expressed enzymes , protective protein / cathepsin A ( PPCA ) , neuraminidase ( Neu 1 ) , and beta glucosidase , were readily taken up and restored lysosomal function in enzyme deficient mouse macrophages . ^^^ The capacity of recombinant PPCA and Neu 1 to clear the lysosomal storage in target cells was assessed in PPCA / mice , a model of galactosialidosis . ^^^

Defect of the EBP as a chaperon for tropoelastin and a component of receptor complex among neuraminidase 1 ( NEU 1 ) and protective protein / cathepsin A ( PPCA ) is suggested responsible for impaired elastogenesis in autosomal recessive beta Gal , PPCA and NEU 1 deficiencies . ^^^ The purpose of this study is to determine effects of GLB 1 , PPCA and NEU 1 gene mutations on elastogenesis in skin fibroblasts . ^^^ Apparently normal elastogenesis and EBP mRNA expression were observed for fibroblasts from Morquio B disease cases with the GLB 1 gene alleles ( W273L / W273L , W273L / R482H and W273L / W509C substitutions , respectively ) , a galactosialidosis case with the PPCA allele ( IVS7+3A / IVS7+3A ) and a sialidosis case with the NEU 1 allele ( V217M / G243R ) as well as normal subject . ^^^ In this study , the W273L substitution in the EBP could impossibly cause the proposed defect of elastogenesis , and the typical PPCA splicing mutation and the V217M / G243R substitutions in the NEU 1 might hardly have effects on elastic fiber formation in the dermal fibroblasts . . ^^^

METHODS : Six wild type ( 2 months postnatal ) ( Neu1+ / + , PPCA+ / + and beta gal+ / + ) mice were used , and Neu 1 , PPCA and beta gal homozygous ( Neu 1 / , PPCA / and beta gal / ) mice at the same age used as control in this experiment . ^^^ RESULTS : There was a similar distributive pattern of Neu 1 , PPCA and betagal in the inner ear . ^^^ A positive staining of PPCA and beta gal was presented in Neu 1 / mice , and as well as Neu 1 and PPCA in beta gal / mice . ^^^ However , the staining of Neu 1 was not presented , and only very weak staining of beta gal in PPCA / mice . ^^^ CONCLUSION : Neu 1 PPCA and beta gal are distributed in the inner ear of mouse , and the three enzymes also form a lysosomal multi enzyme complex in the inner ear . ^^^
