A novel repressor , par 4 , modulates transcription and growth suppression functions of the Wilms ' tumor suppressor WT 1 . ^^^ In this study , we identified par 4 ( for prostate apoptosis response ) as a WT 1 interacting protein that itself functions as a transcriptional repressor . par 4 contains a putative leucine zipper domain and is specifically upregulated during apoptosis of prostate cells ( S . ^^^ The leucine repeat domain of par 4 was shown to interact with the zinc finger DNA binding domain of WT 1 . ^^^ Immunoprecipitation Western blot ( immunoblot ) analyses demonstrated in vivo WT 1 par 4 interactions . par 4 was ubiquitously expressed , and the protein was found in both the nucleus and the cytoplasm . ^^^ Functionally , par 4 inhibited transcription activated by WT 1 , but not by the related protein EGR 1 . ^^^

Par 4 transcriptionally regulates Bcl 2 through a WT 1 binding site on the bcl 2 promoter . ^^^ We investigated the role of the product of the prostate apoptosis response gene 4 ( Par 4 ) and the product of the Wilms ' tumor 1 gene ( WT 1 ) in the regulation of Bcl 2 expression in prostate cancer cell lines . ^^^ The decrease in the Bcl 2 protein and transcript following all trans retinoic acid treatment was accompanied by changes in localization of Par 4 and an induction in the expression of WT 1 protein . ^^^ The involvement of WT 1 in the Par 4 mediated down modulation of Bcl 2 was further defined by blocking endogenous WT 1 expression , which resulted in an increase in Bcl 2 expression . ^^^ Finally , we detected Par 4 and WT 1 proteins binding to a previously identified WT 1 binding site on the bcl 2 promoter both in vitro and in vivo leading to a decrease in transcription from the bcl 2 promoter . ^^^
