We have identified two novel yeast genes , THO 1 and THO 2 , that partially suppress the transcription defects of hpr1Delta mutants by overexpression . ^^^ We show by in vivo transcriptional and recombinational analysis of tho2Delta cells that THO 2 plays a role in RNA polymerase 2 ( RNA pol 2 ) dependent transcription and is required for the stability of DNA repeats , as previously shown for HPR 1 . ^^^ Our work not only identifies a novel yeast gene , THO 2 , with similar function to HPR 1 , but also provides new evidence for transcriptional blocks as a source of recombination . ^^^

A protein complex containing Tho 2 , Hpr 1 , Mft 1 and a novel protein , Thp 2 , connects transcription elongation with mitotic recombination in Saccharomyces cerevisiae . ^^^ We have shown previously that the yeast genes HPR 1 and THO 2 may be keys to the understanding of transcription associated recombination , as they both affect transcription elongation and hyper recombination in a concerted manner . ^^^ Using a yeast strain that has the wild type THO 2 gene replaced by one encoding a His ( 6 ) HA tagged version , we have isolated an oligomeric complex containing four proteins : Tho 2 , Hpr 1 , Mft 1 and a novel protein that we have named Thp 2 . ^^^ We have reciprocally identified a complex containing Hpr 1 , Tho 2 and Mft 1 using anti Mft 1 antibodies in immunoprecipitation experiments . ^^^ The protein complex is mainly nuclear ; therefore , Tho 2 and Hpr 1 are physically associated . ^^^
