A novel cellular protein ( MTBP ) binds to MDM 2 and induces a G 1 arrest that is suppressed by MDM 2 . ^^^ Using a yeast two hybrid screen , we have identified a gene that encodes a novel cellular protein ( MTBP ) that binds to MDM 2 . ^^^ MTBP can induce G ( 1 ) arrest , which in turn can be blocked by MDM 2 . ^^^

Regulation of p 53 and MDM 2 activity by MTBP . p 53 is a critical coordinator of a wide range of stress responses . ^^^ We have discovered that MTBP promotes MDM 2 mediated ubiquitination and degradation of p 53 and also MDM 2 stabilization in an MDM 2 RING finger dependent manner . ^^^ Moreover , using small interfering RNA to down regulate endogenous MTBP in unstressed cells , we have found that MTBP significantly contributes to MDM 2 mediated regulation of p 53 levels and activity . ^^^ Our findings suggest that MTBP differentially regulates the E 3 ubiquitin ligase activity of MDM 2 towards two of its most critical targets ( itself and p 53 ) and in doing so significantly contributes to MDM 2 dependent p 53 homeostasis in unstressed cells . . ^^^
