Surprisingly , we now found that the human homologue APH 1a carrying the equivalent mutation G122D is fully active in yeast co expressing PS 1 , NCT , and PEN 2 . ^^^ Separate overexpression of PS 1 , NCT , or PEN 2 together with APH 1a G122D allowed the formation of heterodimers lacking the other endogenous components . ^^^ Only the combined overexpression of PS 1 and NCT together with APH 1a G122D facilitated the formation of a fully active gamma secretase complex . ^^^ We conclude that cooperative effects may stabilize a trim eric complex of APH 1a G122D together with PS 1 and NCT . ^^^

When we co infected Sf 9 cells with human presenilin 1 ( PS 1 ) , nicastrin , APH 1a , and PEN 2 , a high molecular weight membrane protein complex that contained PS 1 exclusively in its fragment form associated with three other cofactor proteins was reconstituted and recovered in a highly gamma secretase active state in budded virus particles , whereas nonfunctional PS 1 holoproteins massively contaminated the parental Sf 9 cell membranes . ^^^
