Fbw 7 depletion resulted in accumulation of phosphorylated c Jun , stimulation of AP 1 activity , and neuronal apoptosis . ^^^ We report that in neurons the stability of c Jun is regulated by the E 3 ligase SCF ( Fbw 7 ) , which ubiquitinates phosphorylated c Jun and facilitates c Jun degradation . ^^^ SCF ( Fbw 7 ) therefore antagonizes the apoptotic c Jun dependent effector arm of JNK signaling , allowing neurons to tolerate potentially neurotoxic JNK activity . . ^^^

The 5 Jun point mutation allows c Jun to escape GSK 3 dependent recognition and destruction by the Fbw 7 ubiquitin ligase . ^^^ Similar to c Myc , we report here that phosphorylation of c Jun by GSK 3 creates a high affinity binding site for the E 3 ligase Fbw 7 , which targets c Jun for polyubiquitination and proteasomal degradation . ^^^ These findings explain the enhanced stability and oncogenicity of 5 Jun relative to its cellular counterpart and reveal that GSK 3 and Fbw 7 coordinately regulate c Jun and c Myc . . ^^^
