Fbxw 7 contributes to the ubiquitin mediated degradation of cyclin E , c Myc , Aurora A , Notch and c Jun , all of which appear to function as cell cycle promoters and oncogenic proteins . ^^^ In the present study , we examined the abundance of cyclin E , c Myc and Aurora A in seven cancer cell lines , which harbor wild type ( three lines ) or mutant ( four lines ) Fbxw 7 . ^^^ Forced expression of Fbxw 7 reduced the levels of cyclin E , c Myc and Aurora A in the Fbxw 7 mutant cells . ^^^ In contrast , a decrease in the expression of cyclin E , c Myc or Aurora A by RNA interference significantly suppressed the rate of proliferation and anchorage independent growth of the Fbxw 7 mutant cells . ^^^ These findings thus suggest that the loss of Fbxw 7 results in accumulation of cyclin E , c Myc and Aurora A , all of which appear to be required for growth promotion of cancer cells . ^^^

The Fbw 7 tumor suppressor regulates glycogen synthase kinase 3 phosphorylation dependent c Myc protein degradation . ^^^ We show here that Fbw 7 , a component of the SCF ( Fbw 7 ) ubiquitin ligase and a tumor suppressor , promotes proteasome dependent c Myc turnover in vivo and c Myc ubiquitination in vitro . ^^^ Phosphorylation of c Myc on threonine 58 ( T 58 ) by glycogen synthase kinase 3 regulates the binding of Fbw 7 to c Myc as well as Fbw 7 mediated c Myc degradation and ubiquitination . ^^^ T 58 is the most frequent site of c myc mutations in lymphoma cells , and our findings suggest that c Myc activation is one of the key oncogenic consequences of Fbw 7 loss in cancer . ^^^ Because Fbw 7 mediates the degradation of cyclin E , Notch , and c Jun , as well as c Myc , the loss of Fbw 7 is likely to elicit profound effects on cell proliferation during tumorigenesis . . ^^^

Similar to c Myc , we report here that phosphorylation of c Jun by GSK 3 creates a high affinity binding site for the E 3 ligase Fbw 7 , which targets c Jun for polyubiquitination and proteasomal degradation . ^^^ These findings explain the enhanced stability and oncogenicity of 5 Jun relative to its cellular counterpart and reveal that GSK 3 and Fbw 7 coordinately regulate c Jun and c Myc . . ^^^
