However , an inactive form has also been isolated that lacks one of the five subunits , called TFIIIC 110 . ^^^ A model was proposed in which pol 3 transcription might be regulated by the specific induction of TFIIIC 110 , allowing formation of active TFIIIC from the inactive form . ^^^ We have tested this model by transient transfection of HeLa and HEK 293 cells with a vector expressing TFIIIC 110 . ^^^ We have also made stably transfected HeLa cell lines that carry a doxycycline inducible version of the cDNA for TFIIIC 110 . ^^^ We show that the induced TFIIIC 110 enters the nucleus , binds other TFIIIC subunits and is recruited to tRNA and 5S rRNA genes in vivo . ^^^

Cloning and characterization of a TFIIIC 2 subunit ( TFIIIC beta ) whose presence correlates with activation of RNA polymerase 3 mediated transcription by adenovirus E1A expression and serum factors . ^^^ To investigate this possibility , we have cloned and expressed a cDNA encoding the 110 kD subunit ( TFIIIC beta ) . ^^^ Immunoprecipitation studies with anti TFIIIC beta antibodies have confirmed that TFIIIC beta is a bona fide subunit present only in TFIIIC2a , that TFIIIC2a and the general factor TFIIIC 1 are associated in unfractionated extracts , and that previously undetected polypeptides ( potential TFIIIC 1 subunits ) can be isolated in association with TFIIIC2a . ^^^ Studies with antibodies to TFIIIC beta have shown that this is strongly correlated with a selective increase in the cellular concentration of the TFIIIC beta 110 kD subunit and a concomitant rise in the ratio of the active to inactive forms of TFIIIC2 . . ^^^
