PDZK 1 , a multi PDZ domain containing adaptor protein , interacts with various membrane proteins , including the high density lipoprotein ( HDL ) receptor scavenger receptor class B type 1 ( SR BI ) . ^^^ Here we show that PDZK 1 controls in a tissue specific and post transcriptional fashion the expression of SR BI in vivo . ^^^ SR BI protein expression in PDZK 1 knock out ( KO ) mice was reduced by 95 % in the liver , 50 % in the proximal intestine , and not affected in steroidogenic organs ( adrenal , ovary , and testis ) . ^^^ Hepatic expression of SR BII , a minor splice variant with an alternative C terminal cytoplasmic domain , was not affected in PDZK 1 KO mice , suggesting that binding of PDZK 1 to SR BI is required for controlling hepatic SR BI expression . ^^^ The loss of hepatic SR BI was the likely cause of the elevation in plasma total and HDL cholesterol and the increase in HDL particle size in PDZK 1 KO mice , phenotypes similar to those observed in SR BI KO mice . ^^^

Regulation of SR BI mediated high density lipoprotein metabolism by the tissue specific adaptor protein PDZK 1 . ^^^ This review will focus on one of these lipoprotein receptor adaptor pairs , the high density lipoprotein receptor SR BI ( scavenger receptor class B , type 1 ) and its adaptor PDZK 1 . ^^^ RECENT FINDINGS : The PDZ domain containing adaptor protein PDZK 1 has been shown to bind to and control the activity of the high density lipoprotein receptor SR BI via a tissue specific posttranscriptional mechanism . ^^^ Mice deficient in PDZK 1 have elevated plasma cholesterol levels due to the virtually complete hepatic ablation of SR BI , implicating PDZK 1 as a novel regulator of high density lipoprotein metabolism . ^^^

However , it has recently been shown that fibrates can affect HDL metabolism in mouse by significantly decreasing hepatic levels of the HDL receptor scavenger receptor B 1 ( SR BI ) and the PDZ domain containing protein PDZK 1 . ^^^ PDZK 1 is essential for maintaining hepatic SR BI levels . ^^^ Therefore , decreased SR BI might be secondary to decreased PDZK 1 , but the mechanism by which fibrates lower SR BI has not been elucidated . ^^^ Here we show that feeding PDZK 1 deficient mice fenofibrate resulted in the near absence of SR BI in liver , definitively demonstrating that the effect of fenofibrate on SR BI is PDZK 1 independent . ^^^

Regulation of SR BI protein levels by phosphorylation of its associated protein , PDZK 1 . ^^^ Previously , we identified an SR BI associated protein , termed PDZK 1 , from rat liver membrane extracts . ^^^ PDZK 1 contains four PSD 95 / Dlg / ZO 1 ( PDZ ) domains , the first of which in the N terminal region is responsible for the association with SR BI . ^^^ PDZK 1 controls hepatic SR BI expression in a posttranscriptional fashion both in cell culture and in vivo . ^^^ In this study , we demonstrated that the C terminal region of PDZK 1 is crucial for up regulating SR BI protein expression . ^^^
