Wnt signaling was up regulated in normal FLS by transfection with a Wnt 1 expression plasmid and was down regulated in RA FLS by transfection with dominant negative lymphoid enhancer factor 1 ( LEF 1 ) / T cell factor 4 ( TCF 4 ) and secreted Frizzled receptor protein 1 ( sFRP 1 ) expression plasmids . ^^^ Recombinant sFRP 1 and anti Wnt 1 antibody were also administered to RA FLS to block Wnt signaling . ^^^ In a complementary manner , interference with Wnt signaling using anti Wnt 1 antibody , the Wnt antagonist sFRP 1 , or dominant negative vectors that inhibited the transcription factors TCF 4 / LEF 1 blocked the expression of fibronectin by RA FLS and reduced cell survival . ^^^ Anti Wnt 1 antibody and sFRP 1 also blocked the expression of proMMP 3 . ^^^ |

To explore the role of Frp and its relationship to the Wnt signalling pathway in breast cancer , in situ hybridization and immunohistochemical analyses of Frp , Wnt 1 , APC , beta catenin , and its target genes c myc and cyclin D 1 were conducted in 70 specimens of invasive ductal carcinomas of the human breast . ^^^ The level of Wnt 1 was linearly correlated with its downstream target beta catenin ( p < 0 . 05 ) , but was inversely correlated with Frp ( p < 0 . 05 ) , suggesting a possible negative regulatory role of Frp with regard to Wnt 1 . ^^^ |

A secreted frizzled related protein , FrzA , selectively associates with Wnt 1 protein and regulates wnt 1 signaling . ^^^ Here we demonstrate that FrzA , a sFRP that is highly expressed in vascular endothelium and a variety of epithelium , specifically binds to Wnt 1 protein , but not Wnt 5a protein , and modulates Wnt 1 signaling . ^^^ FrzA associated with Wnt 1 either when expressed in the same cell or when soluble FrzA was incubated with Wnt 1 expressing cells . ^^^ FrzA efficiently inhibited the Wnt 1 mediated increase in cytoplasmic ( beta ) catenin levels as well as the Wnt 1 induction of transcription from a Lef / tcf reporter gene . ^^^ The effects of FrzA on ( beta ) catenin levels could be demonstrated when co expressed with Wnt 1 or when individual cells expressing FrzA and Wnt 1 were co cultured . ^^^ |
