| We show that GATA 2 is covalently modified by small ubiquitin like modifier ( SUMO ) 1 and 2 and that PIASy , through its E 3 SUMO ligase activity , preferentially enhances the conjugation of SUMO 2 to GATA 2 . ^^^ We conclude that PIASy enhances the conjugation of SUMO 2 to GATA 2 and that the interaction of PIASy with GATA 2 can modulate GATA mediated ET 1 transcription activity in endothelial cells through a RING like domain independent mechanism . . ^^^ |

| PIASy mediates SUMO 2 conjugation of Topoisomerase 2 on mitotic chromosomes . ^^^ Here we show that the PIASy protein is specifically required for mitotic modification of Topoisomerase 2 by SUMO 2 conjugation in Xenopus egg extracts . ^^^ We observed that PIASy depletion eliminated essentially all chromosomal accumulation of EGFP SUMO 2 conjugated species , suggesting that it is the primary E 3 like factor for mitotic chromosomal substrates of SUMO 2 . ^^^ PIASy dependent SUMO 2 conjugated species were highly concentrated on the inner centromere , and inhibition of PIASy blocked anaphase sister chromatid segregation in egg extracts . ^^^ Taken together , our observations suggest that PIASy is a critical regulator of mitotic SUMO 2 conjugation for Topoisomerase 2 and other chromosomal substrates , and that its activity may have particular relevance for centromeric functions required for proper chromosome segregation . . ^^^ |
