| Caenorhabditis elegans nucleoporins Nup 93 and Nup 205 determine the limit of nuclear pore complex size exclusion in vivo . ^^^ More specifically , the C . elegans homologs of vertebrate Nup 93 and Nup 205 were each found to be required for normal NPC distribution in the nuclear envelope in vivo . ^^^ Depletion of Nup 93 or Nup 205 caused a failure in nuclear exclusion of nonnuclear macromolecules of approximately 70 kDa without preventing active nuclear protein import or the assembly of the nuclear envelope . ^^^ Thus , the contribution to NPC structure of Nup 93 and Nup 205 is essential for establishment of normal NPC function and for cell viability . . ^^^ |

| By postembedding immunoelectron microscopy , we have mapped the positions of several human NPC proteins relative to the NPC core and its associated basket , including Nup 93 , Nup 96 , Nup 98 , Nup 107 , Nup 153 , Nup 205 , and the coiled coil dominated 267 kDa protein Tpr . ^^^ To further assess their contributions to NPC and basket architecture , the genes encoding Nup 93 , Nup 96 , Nup 107 , and Nup 205 were posttranscriptionally silenced by RNA interference ( RNAi ) in HeLa cells , complementing recent RNAi experiments on Nup 153 and Tpr . ^^^ Nup 93 and Nup 205 are other NPC core elements that are important for long term maintenance of NPCs but initially dispensable for the anchoring of Nup 153 and Tpr . ^^^ |
