We have now identified a new member of the Lmo family , designated Lmo 4 , via its interaction with Ldb 1 . ^^^ These characteristics imply that Ldb 1 Lmo4 interaction may function in the T cell developmental programme and that enforced expression of LMO 1 or LMO 2 by chromosomal translocations or transgenesis may displace Lmo 4 from this complex and thereby influence T cell differentiation prior to T cell tumour occurrence . . ^^^ |

Structural basis for the recognition of ldb 1 by the N terminal LIM domains of LMO 2 and LMO 4 . ^^^ LMO 2 and LMO 4 interact with the ubiquitous nuclear adaptor protein ldb1 / NLI / CLIM2 , which associates with the LIM domains of LMO and LIM homeodomain proteins via its LIM interaction domain ( ldb 1 LID ) . ^^^ We report the solution structures of two LMO : ldb 1 complexes ( PDB : 1M3V and 1J2O ) and show that ldb 1 LID binds to the N terminal LIM domain ( LIM 1 ) of LMO 2 and LMO 4 in an extended conformation , contributing a third strand to a beta hairpin in LIM 1 domains . ^^^ |

The LIM only protein , LMO 4 , and the LIM domain binding protein , LDB 1 , expression in squamous cell carcinomas of the oral cavity . ^^^ LMO 4 mRNA was amplified in four of six carcinoma tissues and eight of 12 carcinoma cell lines , and LDB 1 in three carcinoma tissues and 11 cell lines examined . ^^^ Immunoprecipitation studies revealed that LMO 4 and LDB 1 interact with each other in the nuclear milieu of the carcinoma cells indicating the presence of an LMO 4 LDB1 mediated transcription complex . ^^^ Both LMO 4 and LDB 1 proteins were preferentially localised in the nuclei of carcinoma cells at the invasive front and the immunoreactivity was increased in less differentiated carcinoma tissues ( P < 0 . 01 ) . ^^^ |

LMO 4 is comprised primarily of two tandemly repeated LIM domains and interacts with the ubiquitous nuclear adaptor protein ldb 1 . ^^^ This interaction is mediated via the LIM domains of LMO 4 and the LIM interaction domain ( LID ) of ldb 1 . ^^^ An intramolecular complex , termed FLINC 4 , consisting of the two LIM domains from LMO 4 linked to the LID domain of ldb 1 via a flexible linker has been engineered , purified and crystallized . ^^^ |

Tandem LIM domains provide synergistic binding in the LMO 4 : Ldb 1 complex . ^^^ We have determined a high resolution 10 ray crystal structure of LMO 4 , a putative breast oncoprotein , in complex with Ldb 1 LID , providing the first example of a tandem LIM : Ldb 1 LID complex and the first structure of a type B LIM domain . ^^^ The complex possesses a highly modular structure with Ldb 1 LID binding in an extended manner across both LIM domains of LMO 4 . ^^^ |
