A novel locus for Meckel Gruber syndrome , MKS 3 , maps to chromosome 8q24 . ^^^ We report the localisation of a third MKS locus ( MKS 3 ) to chromosome 8q24 in this cohort by a genome wide linkage search using autozygosity mapping . ^^^ Furthermore , a heterozygous SNP in DDEF 1 , a putative candidate gene , suggested that MKS 3 mapped within a 15 cM interval . ^^^ Comparison of the clinical features of MKS 3 linked cases with reports of MKS 1 and MKS 2 linked kindreds suggests that polydactyly ( and possibly encephalocele ) appear less common in MKS 3 linked families . . ^^^

Finally , we have analyzed genes present in the other Meckel syndrome critical regions , MKS 2 and MKS 3 , to determine whether any of the candidate genes for the three MKS loci have similar gene functions or are members of a common biological pathway . . ^^^
