Expression patterns of interleukin 6 ( IL 6 ) , IL 6 receptor ( IL 6R ) , and gp 130 genes in 39 patients with acute myeloid leukemia ( AML ) , in 23 patients with acute lymphoblastic leukemia ( ALL ) , and in 7 patients with acute mixed lineage leukemia ( AMLL ) were studied by quantitative reverse transcriptase polymerase chain reaction . ^^^ Significant correlation was observed between the expression of IL 6R and gp 130 in AML . ^^^ When tested for in vitro response to IL 6 , the leukemic cells from 3 of 7 AML , none of 3 ALL , and both of 2 AMLL patients significantly responded to IL 6 , showing the correlation between the expression levels of IL 6R and gp 130 and the responsiveness of leukemic cells to IL 6 . ^^^ These results showed that quantitation of IL 6R and gp 130 expression by reverse transcriptase polymerase chain reaction is useful for the rapid prediction of the responsiveness of leukemic cells to IL 6 , especially in cases of administration of IL 6 . . ^^^ |

The IL 6 receptor system comprises two functionally different chains : a binding chain ( IL 6R ) and a signal transducing chain ( gp 130 ) . ^^^ The IL 6 / IL 6R complexes associate with gp 130 , induce its dimerization and signal transduction . ^^^ When IL 6 is complexed to IL 6R , two distinct sites of IL 6 are able to bind gp 130 . ^^^ These IL 6RA carried substitutions that increased their affinity with IL 6R and abolished 1 or the 2 sites of interaction with gp 130 . ^^^ The results indicate that the interaction of one gp 130 chain with IL 6R / IL 6R complexes inhibited further the dimerization of gp 130 induced by IL 11 / IL 11R but not its heterodimerization with LIFR . ^^^ |

We analysed the expression of both components of IL 6R , CD 126 the ligand binding protein and CD 130 the signal transducing protein , on plasma cells from MGUS and multiple myeloma ( MM ) cases using flow cytometry . ^^^ CD 130 rather than CD 126 expression is associated with disease activity in multiple myeloma . ^^^ Finally , combining CD 126 and CD 130 expression we found a significant increase of the percentage of CD126+ CD130+ patients at relapse , underlying the crucial role of IL 6 response in the late stage of MM . . ^^^ |
