Distinct functions for WRN and TP 53 in a shared pathway of cellular response to 1 beta D arabinofuranosylcytosine and bleomycin . ^^^ Mutations in the WRN or the TP 53 genes lead to spontaneous genetic instability , an elevated risk of tumor formation , and sensitivity to compounds that interfere with DNA replication , such as camptothecin and DNA interstrand cross linking drugs . ^^^ We investigated the hypothesis that WRN and TP 53 are involved in cellular responses to DNA replication blocking lesions by exposing WRN deficient and TP 53 mutant lymphoblastoid cell lines ( LCLs ) to 1 beta d arabinofuranosylcytosine ( AraC ) and bleomycin . ^^^ Loss of WRN or TP 53 function resulted in induction of apoptosis and lesser proliferative survival in response to AraC and bleomycin . ^^^ WRN and TP 53 operate in a shared DNA damage response pathway , since in cells in which TP 53 was inactivated by SV 40 transformation , no difference in AraC and bleomycin sensitivity was found regardless of WRN status . ^^^ |

Polymorphisms of the TP 53 codon 72 and WRN codon 1367 in individuals from Northern Brazil with gastric adenocarcinoma . ^^^ In the present study , the TP 53 codon 72 and the WRN codon 1367 polymorphisms were investigated in 54 gastric adenocarcinoma patients ( 31 diffuse type and 25 intestinal type ) and 54 controls . ^^^ DNA samples were extracted , and PCR RFLP was utilised for genotyping TP 53 codon 72 and WRN codon 1367 . ^^^ The precise mechanisms of action of the TP 53 and WRN polymorphisms involved in the aetiopathogeny of this disease need further investigation . . ^^^ |

We analyzed one or more single nucleotide polymorphisms ( SNPs ) in each of seven candidate genes ( WRN , TP 53 , NBS 1 , BRCA 1 , BRCA 2 , XRCC 3 and XRCC 4 ) in a study of 250 workers exposed to benzene and 140 controls in China . ^^^ Four SNPs in WRN ( Ex 4 16 G > A , Ex 6 +9 C > T , Ex 20 88 G > T and Ex 26 12 T > G ) , one SNP in TP 53 ( Ex 4 +119 C > G ) and one SNP in BRCA 2 ( Ex 11 +1487 A > G ) were associated with a statistically significant decrease in total white blood cell ( WBC ) counts among exposed workers . ^^^ The SNPs in WRN and TP 53 remained significant after accounting for multiple comparisons . ^^^ These results suggest that genetic polymorphisms in WRN , TP 53 and BRCA 2 that maintain genomic stability impact benzene induced hematotoxicity . . ^^^ |
