To obtain a biochemical marker for Arabidopsis eIF 3 , we cloned the Arabidopsis ortholog of the eIF 3 subunit eIF3b ( PRT 1 ) . eIF3e coimmunoprecipitated with CSN 7 , and eIF3c coimmunoprecipitated with eIF3e , eIF3b , CSN 8 , and CSN 1 . eIF3e directly interacted with CSN 7 and eIF3c . ^^^ Whereas eIF 3 , as detected through eIF3b , localized solely to the cytoplasm , eIF3e , like CSN 7 , was also found in the nucleus . ^^^

Saccharomyces cerevisiae protein Pci8p and human protein eIF3e / Int 6 interact with the eIF 3 core complex by binding to cognate eIF3b subunits . ^^^ We show that both Pci8p and human eIF3e / Int 6 expressed in budding yeast interact with the yeast eIF 3 complex in vivo and in vitro by binding to a discrete segment of its eIF3b subunit Prt1p and that human eIF3e / Int 6 interacts with the human eIF3b segment homologous to the Pci8p binding site of yeast Prt1p . ^^^

We found that mutating the RNP 1 motif in the predicted RRM domain in yeast eukaryotic initiation factor 3 ( eIF 3 ) subunit b / PRT1 ( prt 1 rnp1 ) impairs its direct interactions in vitro with both eIF3a / TIF32 and eIF3j / HCR1 . ^^^ The rnp 1 mutation in PRT 1 confers temperature sensitive translation initiation in vivo and reduces 40S binding of eIF 3 to native preinitiation complexes . ^^^ Several findings indicate that the rnp 1 lesion decreases recruitment of eIF 3 to the 40S subunit by HCR 1 : ( 1 ) rnp 1 strongly impairs the association of HCR 1 with PRT 1 without substantially disrupting the eIF 3 complex ; ( 2 ) rnp 1 impairs the 40S binding of eIF 3 more so than the 40S binding of HCR 1 ; ( 3 ) overexpressing HCR 1 R215I decreases the Ts ( ) phenotype and increases 40S bound eIF 3 in rnp 1 cells ; ( 4 ) the rnp 1 Ts ( ) phenotype is exacerbated by tif 32 Delta6 , which eliminates a binding determinant for HCR 1 in TIF 32 ; and ( 5 ) hcr1Delta impairs 40S binding of eIF 3 in otherwise wild type cells . ^^^ Thus , the PRT 1 RNP1 motif coordinates the functions of HCR 1 and TIF 32 in 40S binding of eIF 3 and is needed for optimal preinitiation complex assembly and AUG recognition in vivo . . ^^^
