Here we describe the association between components of the COP 9 signalosome ( CSN 1 , CSN 7 , and CSN 8 ) and two subunits of eukaryotic translation initiation factor 3 ( eIF 3 ) , eIF3e ( p 48 , known also as INT 6 ) and eIF3c ( p 105 ) . ^^^ To obtain a biochemical marker for Arabidopsis eIF 3 , we cloned the Arabidopsis ortholog of the eIF 3 subunit eIF3b ( PRT 1 ) . eIF3e coimmunoprecipitated with CSN 7 , and eIF3c coimmunoprecipitated with eIF3e , eIF3b , CSN 8 , and CSN 1 . eIF3e directly interacted with CSN 7 and eIF3c . ^^^ Whereas eIF 3 , as detected through eIF3b , localized solely to the cytoplasm , eIF3e , like CSN 7 , was also found in the nucleus . ^^^

The murine int 6 locus , identified as a frequent integration site of mouse mammary tumor viruses , encodes the 48 kDa eIF3e subunit of translation initiation factor eIF 3 . ^^^ Previous studies indicated that the catalytically active core of budding yeast eIF 3 consists of five subunits , all conserved in eukaryotes , but does not contain a protein closely related to eIF3e / Int 6 . ^^^

Moe 1 and spInt 6 , the fission yeast homologues of mammalian translation initiation factor 3 subunits p 66 ( eIF3d ) and p 48 ( eIF3e ) , respectively , are required for stable association of eIF 3 subunits . ^^^ These phenotypes of Deltamoe 1 cells are similar to those reported previously for deletion of the fission yeast int 6 ( + ) gene that encodes the fission yeast homologue of the p48 / Int6 / eIF3e subunit of mammalian eIF 3 . ^^^

The Int 6 gene is a site of mouse mammary tumour virus ( MMTV ) integration in murine tumours and INT 6 protein has been identified independently as a subunit ( eIF3e ) of the eukaryotic translation initiation factor eIF 3 . ^^^ Here , using immunofluorescence microscopy , we show that eIF3e / INT6 is localized in part to the nucleus , while other eIF 3 components are cytoplasmic . ^^^

PCI proteins eIF3e and eIF3m define distinct translation initiation factor 3 complexes . : BACKGROUND : PCI / MPN domain protein complexes comprise the 19S proteasome lid , the COP 9 signalosome ( CSN ) , and eukaryotic translation initiation factor 3 ( eIF 3 ) . ^^^ RESULTS : Here , we report the unexpected finding that fission yeast has two distinct eIF 3 complexes sharing common core subunits , but distinguished by the PCI proteins eIF3e and the novel eIF3m , which was previously annotated as a putative CSN subunit . ^^^ CONCLUSION : We propose that the PCI proteins eIF3e and eIF3m define distinct eIF 3 complexes that may assist in the translation of different sets of mRNAs . . ^^^

Translation initiation factor eIF4G 1 binds to eIF 3 through the eIF3e subunit . eIF 3 in mammals is the largest translation initiation factor ( approximately 800 kDa ) and is composed of 13 nonidentical subunits designated eIF3a m . ^^^ However , binding of partially proteolyzed HeLa eIF 3 to the eIF 3 binding domain of human eIF4G 1 , followed by high throughput analysis of mass spectrometric data with a novel peptide matching algorithm , identified a single subunit , eIF3e ( p48 / Int 6 ) . ^^^ In addition , recombinant FLAG eIF3e specifically competed with HeLa eIF 3 for binding to eIF4G in vitro . ^^^ These data suggest a specific , direct , and functional interaction of eIF3e with eIF4G during the process of cap dependent translation initiation , although they do not rule out participation of other eIF 3 subunits . . ^^^

Here we report the cloning and characterization of human cDNAs encoding the major RNA binding subunit , eIF 3 p66 , and two additional subunits , eIF 3 p47 and eIF 3 p40 . ^^^ The N terminal regions of human eIF 3 p40 and eIF 3 p47 are related to each other and to 17 other eukaryotic proteins , including murine Mov 34 , a subunit of the 26 S proteasome . ^^^ Phylogenetic analyses of the 19 related protein sequences , called the Mov 34 family , distinguish five major subgroups , where eIF 3 p40 , eIF 3 p47 , and Mov 34 are each found in a different subgroup . ^^^

We used a yeast two hybrid screening strategy and identified eukaryotic initiation factor 3 p 47 protein ( eIF 3 p47 ) as an interacting partner of caspase processed C terminal kinase domain of CDK 11 ( CDK 11 ( p 46 ) ) . ^^^ We demonstrate that the eIF 3 p47 can interact with CDK 11 in vitro and in vivo , and the interaction can be strengthened by stimulation of apoptosis . ^^^ EIF 3 p47 contains a Mov34 / JAB domain and appears to interact with CDK 11 ( p 46 ) through this motif . ^^^ We show in vitro that the caspase processed CDK 11 ( p 46 ) can phosphorylate eIF 3 p47 at a specific serine residue ( Ser ( 46 ) ) and that eIF 3 p47 is phosphorylated in vivo during apoptosis . ^^^
