Trif recruited the kinases receptor interacting protein ( RIP ) 1 and RIP 3 through its RIP homotypic interaction motif . ^^^ In the absence of RIP 1 , TLR 3 mediated signals activating NF kappa B , but not the kinase JNK or interferon beta , were abolished , suggesting that RIP 1 mediates Trif induced NF kappa B activation . ^^^ In contrast , the presence of RIP 3 negatively regulated the Trif RIP 1 induced NF kappa B pathway . ^^^

Using macrophages in which receptor interacting protein 1 ( RIP 1 ) was knocked down by small interfering RNA , and macrophages isolated from Toll / interleukin 1 receptor domain containing adaptor inducing IFN beta ( TRIF ) deficient mice , we found that TRIF and RIP 1 function upstream of ROS production in LPS + Z VAD treated macrophages . ^^^ This paper reveals that TRIF , RIP 1 , and ROS production , as well as PARP activation , are involved in inducing autophagy , which contributes to caspase independent macrophage cell death . . ^^^
