In this report , we have shown that cIAP 2 specifically mediated ubiquitination and proteasome dependent degradation of TRAF 1 . ^^^ To identify the sites for cIAP 2 mediated ubiquitination of TRAF 1 , we used high pressure liquid chromatography coupled with tandem mass spectrometry . ^^^ Mutation of Lys 185 and Lys 193 to Arg almost completely blocked cIAP 2 mediated ubiquitination of TRAF 1 , indicating that they are the major , if not the only , sites of TRAF 1 ubiquitination . ^^^ Our data suggest that cIAP 2 may regulate the turnover of TRAF 1 by adding polyubiquitin chains on Lys 185 or Lys 193 following its recruitment to TNF R signaling complexes . . ^^^
