RhoE function is regulated by ROCK 1 mediated phosphorylation . ^^^ The Rho GTPase family member RhoE regulates actin filaments partly by binding to and inhibiting ROCK 1 , a serine / threonine kinase that induces actomyosin contractility . ^^^ Here , we show that ROCK 1 can phosphorylate multiple residues on RhoE in vitro . ^^^ In cells , ROCK 1 phosphorylated RhoE localizes in the cytosol , whereas unphosphorylated RhoE is primarily associated with membranes . ^^^ Phosphorylation has no effect on RhoE binding to ROCK 1 , but instead increases RhoE protein stability . ^^^ |

In part , this is due to RhoE interaction with the RhoA effector ROCK 1 , a serine / threonine kinase that regulates the formation and contractility of stress fibers . ^^^ RhoE binding inhibits ROCK 1 from phosphorylating its downstream target myosin light chain phosphatase , thus increasing the activity of the phosphatase to dephosphorylate myosin 2 , which results in reduced actomyosin contractility . ^^^ RhoE itself is phosphorylated by ROCK 1 , and this may enhance RhoE regulation of ROCK 1 function . ^^^ |
