| Integrin linked protein kinase regulates fibronectin matrix assembly , E cadherin expression , and tumorigenicity . ^^^ We report here that overexpression of integrin linked kinase ( ILK ) , a newly identified serine / threonine kinase that binds to the integrin beta 1 cytoplasmic domain , dramatically stimulated Fn matrix assembly in epithelial cells . ^^^ The integrin linked kinase activity is involved in transducing signals leading to the up regulation of Fn matrix assembly , as overexpression of a kinase inactive ILK mutant failed to enhance the matrix assembly . ^^^ These results identify ILK as an important regulator of pericellular Fn matrix assembly , and suggest a novel critical role of this integrin linked kinase in cell growth , cell survival , and tumorigenesis . . ^^^ |

| C . elegans PAT 4 / ILK functions as an adaptor protein within integrin adhesion complexes . ^^^ BACKGROUND : Mammalian integrin linked kinase ( ILK ) was identified in a yeast two hybrid screen for proteins binding the integrin beta ( 1 ) subunit cytoplasmic domain . ^^^ ILK has been implicated in integrin mediated signaling and is also an adaptor within integrin associated cytoskeletal complexes . ^^^ Conversely , PAT 4 / ILK requires the ECM component UNC 52 / perlecan , the transmembrane protein integrin , and the novel cytoplasmic attachment protein UNC 112 to be properly recruited to nascent attachments . ^^^ CONCLUSIONS : Our data strengthens the emerging view that ILK functions primarily as an adaptor protein within integrin adhesion complexes and identifies UNC 112 as a new ILK binding partner . . ^^^ |

| Integrin linked kinase ( ILK ) is an integrin binding cytoplasmic protein that has been implicated in regulating numerous cellular processes and fibronectin ( Fn ) deposition through mediated integrin , but the expression and significance of ILK in the aging kidney have not yet been reported . ^^^ We report here that mRNA and protein expression of ILK increased in primary cultured mesangial and tubular epithelial cells , and normal and unilateral ureteral obstructed kidney tissues in 28 month old rats but not in 3 month old rats , moreover , accompanied by the over expression of Fn and integrin beta 1 in the aging kidney , by means of Northern blot , Western blot , and immunofluorescent double staining immunohistochemistry . ^^^ |

| We have investigated the effects of high glucose and angiotensin 2 on the expression of integrin alpha 3 and beta 1 , and whether these changes are associated with integrin linked kinase ( ILK ) in cultured mouse podocytes . ^^^ Integrin beta 1 and ILK mRNA expression and protein production were rapidly up regulated in a dose dependent manner by high glucose and angiotensin 2 stimulation . ^^^ Angiotensin 2 induced integrin beta 1 and ILK mRNA expression was significantly inhibited by treatment with losartan ( 100 muM ) . ^^^ In addition , the up regulation of ILK synthesis induced by these stimuli was related to beta 1 integrin synthesis and increased ILK kinase activity . ^^^ Thus , high glucose and angiotensin 2 may affect the regulation of the integrin ILK system in podocytes ; this system may therefore play a role in the pathogenesis of diabetic nephropathy and other renal diseases affecting podocytes . . ^^^ |
