TNF , TRAF 2 , and ultraviolet ( UV ) light , which in part uses the TNF receptor signaling pathway , all increased GCKR activity . ^^^ A TRAF 2 mutant , which inhibits both TRAF 2 induced NF kappaB and SAPK activation , blocked TNF induced GCKR activation . ^^^ Finally , interference with GCKR expression impeded TRAF 2 and TNF induced SAPK activation but not that of NF kappaB . ^^^ This suggests a divergence in the TNF signaling pathway that leads to SAPK and NF kappaB activation , which is located downstream of TRAF 2 but upstream of GCKR . . ^^^

Tumor necrosis factor ( TNF ) induced germinal center kinase related ( GCKR ) and stress activated protein kinase ( SAPK ) activation depends upon the E2 / E3 complex Ubc 13 Uev1A / TNF receptor associated factor 2 ( TRAF 2 ) . ^^^ Although the TRAF 2 TRAF domain binds ASK 1 , GCK , and the highly related kinase GCKR , the RING finger domain is needed for their activation . ^^^ Here , we report that TNF activates GCKR and the SAPK pathway in a manner that depends upon TRAF 2 and Ubc 13 , a member along with Uev1A of a dimeric ubiquitin conjugating enzyme complex . ^^^ Interference with Ubc 13 function or expression inhibits both TNF and TRAF 2 mediated GCKR and SAPK activation , but has a minimal effect on ASK 1 activation . ^^^ TNF signaling leads to TRAF 2 polyubiquitination and oligomerization and to the oligomerization , ubiquitination , and activation of GCKR , all of which are sensitive to the disruption of Ubc 13 function . ^^^
