TNF alpha stimulates the JNK , ERK , and p 38 mitogen activated protein kinases and the NF kappaB pathway by recruiting RIP 1 and TRAF 2 to the TNF receptor 1 . ^^^ Here we showed that Tpl 2 activation by TNF alpha signals depends on the integrity of the Tpl 2 interacting proteins RIP 1 and TRAF 2 , which are required for the engagement of the ERK mitogen activated protein kinase pathway . ^^^ However , neither RIP 1 nor TRAF 2 overexpression was sufficient to activate Tpl 2 and ERK . ^^^ These data therefore dissect the TNF receptor 1 proximal events that regulate Tpl 2 and ERK and highlight a role for RIP 1 , TRAF 2 , and Syk in this pathway . . ^^^ |

We reveal that receptor interacting protein 1 ( RIP 1 ) and tumor necrosis factor receptor associated factor 2 ( TRAF 2 ) , are upstream of JNK in PARP 1 hyperactivated cells , because PARP 1 induced JNK activation was attenuated in RIP 1 / and TRAF 2 / mouse embryonic fibroblast cells . ^^^ Consistently , knockouts of RIP 1 and TRAF 2 caused a resistance to PARP 1 induced cell death . ^^^ Therefore , our study uncovers that RIP 1 , TRAF 2 , and JNK comprise a pathway to mediate the signaling from PARP 1 overactivation to mitochondrial dysfunction . . ^^^ |
