Regulatory mechanisms of TRAF 2 mediated signal transduction by Bcl 10 , a MALT lymphoma associated protein . ^^^ To elucidate the function of Bcl 10 , recently cloned as an apoptosis associated gene mutated in MALT lymphoma , we identified its binding partner TRAF 2 , which mediates signaling via tumor necrosis factor receptors . ^^^ In mammalian cells , low levels of Bcl 10 expression promoted the binding of TRAF 2 and c IAPs . ^^^ Overexpressed Bcl 10 reduced c Jun N terminal kinase activation and induced nuclear factor kappaB activation downstream of TRAF 2 . ^^^ The phenotype of Bcl 10 transgenic mice was similar to that of TRAF 2 deficient mice reported previously , indicating that excessive expression of Bcl 10 might deplete the TRAF 2 function . ^^^

Previously , we have demonstrated that Bcl 10 potentially interacts with the other apoptosis regulator , TNF receptor associated factor 2 ( TRAF 2 ) and inhibitor of apoptosis proteins ( cIAPs ) . ^^^ Interchangeable binding of Bcl 10 to TRAF 2 and cIAPs regulates apoptosis signaling . ^^^ The present results showed that the complex formation of these molecules was regulated by phosphorylation of Bcl 10 , that is , phosphorylation of Bcl 10 resulted in binding of Bcl 10 to cIAPs and the dissociation of it from TRAF 2 . ^^^ These findings indicate that Bcl 10 is a mediator of apoptosis signaling , by switching over binding to cIAPs from TRAF 2 through the events of Bcl 10 phosphorylation . . ^^^

RESULTS : Multiple survival factor genes , including cellular inhibitor of apoptosis protein ( c IAP 1 ) , c IAP 2 , TNF receptor associated factor 1 ( TRAF 1 ) , TRAF 2 , B cell leukemia / lymphoma 2 ( Bcl 2 ) , Bcl 10 , A 1 , and cellular Fas associated death domain ( FADD ) like interleukin 1beta converting enzyme like inhibitory protein ( c FLIP ) , were expressed in basal conditions in both cultured RPE cells and RPE cells in situ , whereas survivin was expressed only by cultured cells . ^^^ TRAF 1 , c FLIP , and to a lesser extent c IAP 2 protein levels were increased by TNF alpha in a time dependent manner , whereas c IAP 1 , survivin , Bcl 10 ( L ) , and TRAF 2 protein levels were not influenced by TNF alpha treatment at any time point tested . ^^^
