| We show here that RNA helicase A ( RHA ) protein links BRCA 1 to the holoenzyme complex . ^^^ The region of BRCA 1 which interacts with RHA and , thus , the holoenzyme complex , corresponds to subregions of the BRCT domain of BRCA 1 ( ref . 9 ) . ^^^ This interaction was shown to occur in yeast nuclei , and expression in human cells of a truncated RHA molecule which retains binding to BRCA 1 inhibited transcriptional activation mediated by the BRCA 1 carboxy terminus . ^^^ |

| Utilizing a previously described interaction between BRCA 1 and RNA helicase A ( RHA ) , we have developed a dominant negative approach to block BRCA 1 function in human breast epithelial cells . ^^^ Overexpression of a truncated RHA peptide that can bind to the BRCA 1 carboxy terminus prevents normal BRCA 1 function , such as BRCA 1 association with nuclear foci following DNA damage . ^^^ Furthermore , inhibition of BRCA 1 via overexpressing the RHA fragment coincides with a reduction in PARP 1 protein expression , suggesting a possible mechanism for BRCA 1 in the maintenance of genomic integrity . . ^^^ |
