Receptor interacting protein 140 ( RIP 140 ) , a previously identified putative ligand dependent coactivator of nuclear hormone receptors , was isolated by yeast two hybrid cloning as a factor that interacts with peroxisome proliferator activated receptor alpha ( PPARalpha ) . 0.64752981^^^

Using peroxisome proliferator activated receptor alpha ( PPAR alpha ) as bait in a yeast two hybrid screening , we have isolated nuclear factor RIP 140 whose function in receptor activation is unclear . ^^^ We now report a detailed characterization of RIP 140 action with a focus on the retinoid 10 receptor ( RXR ) heterodimeric receptors PPAR and thyroid hormone receptor ( TR ) . ^^^ We show that putative PPAR ligands enhance the interaction of RIP 140 with the rat PPAR subtypes alpha and gamma in solution but not with PPAR / RXR heterodimers on DNA . ^^^

Further studies demonstrated that the PPARalpha ligand 8 ( S ) hydroxyeicosatetraenoic acid strongly promotes the interaction of PPARalpha with the co activator RIP 140 but decreases the interaction of PPARalpha with the co repressor SMRT . ^^^ In contrast , TAK 1 interacts with RIP 140 but not with SMRT and competes with PPARalpha for RIP 140 binding . ^^^
