Human 4 1BB ( CD 137 ) signals are mediated by TRAF 2 and activate nuclear factor kappa B . ^^^ We found that tumor necrosis factor receptor associated factor ( TRAF ) 1 , TRAF 2 , and TRAF 3 , all interacted with the cytoplasmic domain of 4 1BB . ^^^ Mutation analysis showed that TRAF 1 , TRAF 2 , and TRAF 3 were associated with one of two runs of acidic residues found in the cytoplasmic domain of 4 1BB . ^^^ In addition , 4 1BB cross linking with TCR signal in Jurkat cells and overexpression of 4 1BB in 293 cells were able to induce activation of the nuclear factor kappa B ( NF kappa B ) . 4 1BB mediated NF kappa B activation was inhibited by a dominant negative TRAF 2 or NF kappa B inducing kinase ( NIK ) . ^^^

CD 28 independent , TRAF 2 dependent costimulation of resting T cells by 4 1BB ligand . 4 1BB ligand ( 4 1BBL ) is a member of the tumor necrosis factor ( TNF ) family expressed on activated antigen presenting cells . ^^^ We find that TNF receptor associated factor ( TRAF ) 1 or TRAF 2 associate with a glutathione S transferase 4 1BB cytoplasmic domain fusion protein in vitro . ^^^ In T cells , we find that association of TRAF 1 and TRAF 2 with 4 1BB requires 4 1BB cross linking . ^^^ In support of a functional role for TRAF 2 in 4 1BB signaling , we find that resting T cells isolated from TRAF 2 deficient mice or from mice expressing a dominant negative form of TRAF 2 fail to augment IL 2 production in response to soluble 4 1BBL . ^^^ Thus 4 1BB , via the TRAF 2 molecule , can provide CD 28 independent costimulatory signals to resting T cells . . ^^^
