Although the 10 linked immunodeficiencies 10 linked agammaglobulinemia ( XLA ) , 10 linked severe combined immunodeficiency ( XSCID ) , Wiskott Aldrich syndrome ( WAS ) , 10 linked lymphoproliferative syndrome and 10 linked hyper IgM syndrome have been mapped to loci distributed throughout the 10 chromosome , they have several features in common that suggest that they might be members of a gene family : ( 1 ) all are maintained in the population at approximately the same gene frequency ; ( 2 ) expression of each defect is limited to the hematopoietic system ; ( 3 ) atypical forms of each disorder have been described ; and ( 4 ) obligate carriers of these disorders are normal by all immunologic criteria . ^^^ The failure of carriers of XLA , XSCID , and WAS to show signs of their gene defects can be explained by the preferential use of the normal , nonmutant 10 as the active 10 in the cell lineages affected by the gene defects . ^^^ These three disorders also share an additional feature ; in boys with XLA , XSCID , or WAS there is asynchronous expression of cell surface markers of differentiation or activation . ^^^

Epstein Barr virus transformed B lymphoblastoid cell lines ( EBV transformed LCL ) from three patients with 10 linked agammaglobulinaemia ( XLA ) , six patients with Wiskott Aldrich Syndrome ( WAS ) , and seven normal donors , were tested for growth and differentiation in response to human recombinant IL 4 , a commercially available , low molecular weight B cell growth factor ( BCGFlow ) , and B cell differentiation factor ( BCDF ) secreted by the T 24 cell line , now known to be IL 6 . ^^^ Proliferation ( 3H TdR uptake ) by EBV transformed LCL from both XLA and WAS patients in response to BCGFlow was similar to that obtained with the normal cell lines . ^^^ In addition , three normal and three WAS , but none of the XLA EBV transformed LCL , proliferated a little in response to IL 4 . ^^^ The lack of detectable IgG secretion by the XLA lines was probably due to an absence of precommitted IgG B cell precursors transformed by EBV rather than an intrinsic inability to respond to BCGF and BCDF . ^^^ These results suggest that the abnormal 10 linked genes responsible for XLA and WAS do not interfere with B cell responses to B cell growth and differentiation factors . . ^^^
