| TopBP 1 recruits Brg1 / Brm to repress E2F1 induced apoptosis , a novel pRb independent and E2F1 specific control for cell survival . ^^^ Through its BRCT domain , TopBP 1 interacts with and represses exclusively E2F1 but not other E2F factors . ^^^ TopBP 1 recruits Brg1 / Brm , a central component of the SWI / SNF chromatin remodeling complex , to E2F1 responsive promoters and represses the activities of E2F1 , but not E2F2 or E2F3 . ^^^ Interestingly , TopBP 1 is induced by E2F and interacts with E2F1 during G1 / S transition . ^^^ Thus , TopBP 1 functions as a critical modulator and serves as a negative feedback regulator of E2F1 by inhibiting E2F1 dependent apoptosis during G1 / S transition as well as DNA damage to promote cell survival . . ^^^ |

| In this study , we showed that the transcription of human MCM 10 and TopBP 1 is activated by transcription factors E2F1 3 , but not by factors E2F4 7 . ^^^ Analysis of various MCM 10 and TopBP 1 promoter constructs showed that an E2F responsive sequence in the vicinity of the transcription initiation site is necessary for the E2F1 induced activation of MCM 10 and TopBP 1 gene transcription , which is further suppressed by pRb . ^^^ Although E2F1 was stabilized by ultraviolet ( UV ) irradiation , the mRNA expression level of TopBP 1 was suppressed in HCT 116 human diploid colon cancer cells . ^^^ |
