| Chk 2 activates E2F 1 in response to DNA damage . ^^^ We report that checkpoint kinase 2 ( Chk 2 ) regulates E2F 1 activity in response to the DNA damaging agent etoposide . ^^^ A Chk 2 consensus phosphorylation site in E2F 1 is phosphorylated in response to DNA damage , resulting in protein stabilization , increased half life , transcriptional activation and localization of phosphorylated E2F 1 to discrete nuclear structures . ^^^ Expression of a dominant negative Chk 2 mutant blocks induction of E2F 1 and prevents E2F 1 dependent apoptosis . ^^^ Moreover , E2F 1 is resistant to induction by etoposide in tumour cells expressing mutant chk 2 . ^^^ |

| Here we define a transcription mediated pathway in which deregulation of E2F1 by ectopic E2F expression or Rb inactivation by E 7 of human papillomavirus type 16 signals apoptosis by inducing the expression of Chk 2 , a component of the DNA damage response . ^^^ However , we found that the key commitment step in apoptosis induction is the ability of E2F1 , and not E2F2 , to upregulate Chk 2 expression . ^^^ Our results suggest that E2F1 plays a central role in signaling disturbances in the Rb growth control pathway and , by upregulation of Chk 2 , may sensitize cells to undergo apoptosis . . ^^^ |
