Here we report that both the acidic transcription activation domain mapping to the unique C terminus of chicken c Rel and the F 9 cell specific activation region common to both 5 Rel and c Rel interact with the TATA binding protein ( TBP ) and transcription factor IIB ( TFIIB ) in vitro and in vivo . ^^^ Combined with the observation that the mouse c Rel and human RelA proteins also interact with TBP and TFIIB in vitro , these results suggest that association with basal transcription factors is important for the transcriptional activities of Rel family proteins . . ^^^

Chromatin immunoprecipitation analyses demonstrated that T cell activation triggers in vivo binding of endogenous c Rel , c Jun , C / EBP beta , and HMG IC to the bfl 1 regulatory region , coincident with selective recruitment of coactivators TAFII 250 and p 300 , SWI / SNF chromatin remodeling factor component BRG 1 , and basal transcription factors TATA binding protein ( TBP ) and TFIIB , as well as hyperacetylation of histones H 3 and H 4 . ^^^

Glutathione S transferase pull down assays showed that the Rel homology domain of p 65 is important for binding to TFIIB . ^^^ Molecular modelling , together with the generation of specific point mutants , revealed that residues 41 R and 42 S in the Rel homology domain of p 65 facilitate the interaction with TFIIB . ^^^
