The interaction of the vitamin D receptor ( VDR ) with transcription factor IIB ( TFIIB ) represents a potential physical link between the VDR DNA complex and the transcription preinitiation complex . ^^^ However , the functional relevance of the VDR TFIIB interaction in vitamin D mediated transcription is not well understood . ^^^ In the present study , we used site directed mutagenesis to demonstrate that the structural integrity of the amino terminal zinc finger of TFIIB is essential for VDR TFIIB complex formation . ^^^ Altering the putative zinc coordinating residues ( C 15 , C 34 , C 37 , or H 18 ) to serines abolished TFIIB interaction with the VDR as assessed in a yeast two hybrid system and by in vitro protein interaction assays . ^^^ A mutant N terminal domain [ N TFIIB ( C34S : C37S ) ] that does not interact with VDR had no impact on 1 , 25 ( OH ) 2D3 induced transcription . ^^^ |

Additional probing of protein protein interactions has revealed that VDR also binds to basal transcription factor IIB ( TFIIB ) and , in the presence of 1 , 25 ( OH ) 2D3 , an RXR VDR TFIIB ternary complex can be created in solution . ^^^ Because the AF 2 domains participate neither in VDR RXR heterodimerization nor in TFIIB association , it is hypothesized that they contact , in a ligand dependent fashion , transcriptional coactivators such as those of the steroid receptor coactivator family , constituting yet a third protein protein interaction for VDR . ^^^ The above events , including bridging by coactivators to the TATA binding protein and associated factors , may position VDR such that it is able to attract TFIIB and the balance of the RNA polymerase 2 transcription machinery , culminating in repeated transcriptional initiation of VDRE containing , vitamin D target genes . ^^^ |
