Structure function analysis of human Spt 4 : evidence that hSpt 4 and hSpt 5 exert their roles in transcriptional elongation as parts of the DSIF complex . ^^^ RESULTS : Immunodepletion of hSpt 5 from HeLa nuclear extracts resulted in the efficient co depletion of hSpt 4 . ^^^ Using DSIF depleted nuclear extracts and a series of Spt 4 mutants , we examined the amino acid sequence of hSpt 4 which was important for hSpt 5 binding and for transcriptional repression and activation by DSIF . ^^^ Unexpectedly , the zinc finger of hSpt 4 , which is critical for the yeast counterpart to function in vivo , was dispensable for hSpt 5 binding and for transcriptional regulation in vitro . ^^^ CONCLUSION : These and other results suggest : ( 1 ) that the central region of hSpt 4 is necessary and sufficient for its function in vitro and ( 2 ) that there is no free hSpt 4 or hSpt 5 in cells . ^^^

To study gene regulation mediated by chromatin in mammals , we isolated the human ( SUPT4H ) and murine ( Supt4h ) counterparts of the yeast gene encoding SPT 4 ; the product of this gene presumably interacts with the products of the mammalian homologues ( which we have also cloned ) of yeast SPT 5 and SPT 6 , thereby modulating chromatin formation and activity . ^^^
