| Direct binding of the signal transducing adaptor Grb 2 facilitates down regulation of the cyclin dependent kinase inhibitor p27Kip1 . ^^^ Here we show that Grb 2 and Grb 3 3 , the molecules functioning as an adaptor in the signal transduction pathway , specifically and directly bind to p 27 in the cytoplasm and participate in the regulation of p 27 . ^^^ In living cells , enforcement of the cytoplasmic localization of p 27 , either by artificial manipulation of the nuclear / cytoplasmic transport signal sequence or by coexpression of ectopic Jab1 / CSN5 , markedly enhances the stable interaction between p 27 and Grb 2 . ^^^ Overexpression of Grb 2 accelerates Jab1 / CSN5 mediated degradation of p 27 , while Grb 3 3 expression suppresses it . ^^^ A p 27 mutant unable to bind to Grb 2 is transported into the cytoplasm in cells ectopically expressing Jab1 / CSN5 but is refractory to the subsequent degradation . ^^^ |

| Kip1 inhibition of GRB 2 SOS formation can regulate Ras activation . p 27 ( Kip 1 ) ( p 27 ) is often inappropriately downregulated in aggressive human cancers . ^^^ A yeast two hybrid screen identified growth factor receptor bound protein 2 ( GRB 2 ) as a p 27 binding partner . ^^^ We now demonstrate that p 27 can inhibit GRB 2 function by blocking its association with the guanine nucleotide exchange factor SOS . ^^^ Endogenous p 27 is rapidly exported from the nucleus to the cytoplasm in response to mitogen stimulation , where it binds GRB 2 concomitant with a decrease in GRB 2 associated SOS . ^^^ As predicted , mitogen stimulated p 27 ( / ) cells maintained their GRB 2 SOS complexes for significantly longer . ^^^ |
