| Lck and p59Fyn regulate CD 28 binding to phosphatidylinositol 3 kinase , growth factor receptor bound protein GRB 2 , and T cell specific protein tyrosine kinase ITK : implications for T cell costimulation . ^^^ CD 28 interacts with three intracellular proteins phosphatidylinositol 3 kinase ( PI 3 kinase ) , T cell specific protein tyrosine kinase ITK ( formerly TSK or EMT ) , and the complex between growth factor receptor bound protein 2 and son of sevenless guanine nucleotide exchange protein ( GRB 2 SOS ) . ^^^ PI 3 kinase and GRB 2 bind to the CD 28 phosphotyrosine based Tyr Met Asn Met motif by means of intrinsic Src homology 2 ( SH 2 ) domains . ^^^ The requirement for tyrosine phosphorylation of the Tyr Met Asn Met motif for SH 2 domain binding implicates an intervening protein tyrosine kinase in the recruitment of PI 3 kinase and GRB 2 by CD 28 . ^^^ In this study , we demonstrate in coexpression studies that p56Lck and p59Fyn phosphorylate CD 28 primarily at Tyr 191 of the Tyr Met Asn Met motif , inducing a 3 to 8 fold increase in p 85 ( subunit of PI 3 kinase ) and GRB 2 SH 2 binding to CD 28 . ^^^ |

| The YMNM motif that exists in the CD 28 cytoplasmic domain is a known binding site for phosphatidylinositol 3 kinase ( PI 3 K ) and Grb 2 . ^^^ ICOS possesses the YMFM motif in the corresponding region of CD 28 that binds PI 3 K but not Grb 2 . ^^^ Further analysis demonstrated that Grb 2 binding to ICOS was sufficient to activate the NFAT / AP 1 site in the IL 2 promoter and that the cytoplasmic domain of CD 28 outside of the YMNM motif is required for activation of the CD28RE / AP 1 and NF kappaB sites . ^^^ Together , these observations lead us to believe that the difference of a single amino acid , which affects Grb 2 binding ability , may define a functional difference between the CD 28 and ICOS mediated costimulatory signals . . ^^^ |

| Grb 2 and Gads exhibit different interactions with CD 28 and play distinct roles in CD 28 mediated costimulation . ^^^ CD 28 binds both PI3K and Grb 2 , whereas ICOS binds only PI3K . ^^^ We have generated an ICOS mutant , which can bind Grb 2 by replacement of its PI3K binding motif YMFM with the CD 28 YMNM motif , and shown that it induces significant activation of the IL 2 promoter . ^^^ In this study , we show that Gads , but not Grb 2 , is essential for CD 28 mediated NF kappaB activation , and its binding to CD 28 requires the whole CD 28 cytoplasmic domain in addition to the YMNM motif . ^^^ Mutagenesis experiments have indicated that mutations in the N terminal and / or C terminal PXXP motif ( s ) of CD 28 significantly reduce their association with Gads , whereas their associations with Grb 2 are maintained . ^^^ |
