Crosstalk between ARF 6 and protein kinase Calpha in Fc ( gamma ) RI mediated activation of phospholipase D 1 . ^^^ Here , we show that immune complex aggregation of Fc ( gamma ) RI stimulates the association of phospholipase D 1 with ARF 6 and protein kinase Calpha . ^^^ Moreover , treatment of cells with butan 1 ol or phospholipase D 1 antisense oligonucleotides inhibits translocation of PKCdelta , epsilon , and zeta but had no effect on the association of PKCalpha or ARF 6 with phospholipase D 1 . ^^^ These data indicate that association with ARF 6 and PKCalpha plays a role in coupling Fc ( gamma ) RI to phospholipase D 1 activation and that PLD 1 lies upstream of all Fc ( gamma ) RI mediated PKC activity . . ^^^

Using a mutant of ARF 6 specifically impaired for PLD 1 stimulation , we find that ARF 6 is functionally linked to phospholipase D ( PLD ) 1 in the exocytotic machinery . ^^^ Finally , we show that ARNO , ARF 6 , and PLD 1 colocalize at sites of exocytosis , and we demonstrate direct interaction between ARF 6 and PLD 1 in stimulated cells . ^^^ Together , these results provide the first direct evidence that ARF 6 plays a role in calcium regulated exocytosis in neuroendocrine cells , and suggest that ARF 6 stimulated PLD 1 activation at the plasma membrane and consequent changes in membrane phospholipid composition are critical for formation of the exocytotic fusion pore . . ^^^
