Chloroform induction of ornithine decarboxylase antizyme ( ODC AZ ) in male rat liver . ^^^ ODC AZ was determined by measuring the inhibition of isolated ODC activity as described by Hayashi and Fujita and modified in our laboratory to use kidney ODC . ^^^ Chloroform induced ODC AZ activity in males at 3 and 7 d ( 26 % and 37 % inhibition of the ODC activity in the incubation medium , respectively ) . ^^^ While females exhibited a similar decline in ODC activity after repeated doses , ODC AZ was not induced . ^^^ Thus , it would appear that daily exposure of rats to chloroform results in a refractoriness of its induction of ODC activity accompanied by an induction of the ODC AZ in males . ^^^ |

Recent studies have provided convincing evidence to add to a number of earlier observations suggesting that the rapid intracellular degradation of mammalian ornithine decarboxylase ( ODC ) is further accelerated by the action of ornithine decarboxylase antizyme ( ODC Az ) , a polyamine induced protein . ^^^ However , the mechanism whereby ODC Az exerts its effect in this proteolytic process is mostly unknown . ^^^ Here , by using reticulocyte lysate based synthesis and degradation systems , we demonstrate that interaction of ODC Az with ODC results in two related outcomes : ( a ) ODC is inactivated as a result of its monomerization , and ( b ) ODC degradation is dramatically accelerated . ^^^ While ODC inactivation requires the integrity of the ODC Az binding site of ODC and the ODC binding site of ODC Az , acceleration in ODC degradation also requires the previously characterized carboxyl terminal destabilizing segment of ODC and a specific segment of ODC Az that may be functionally distinct from that required for ODC binding . ^^^ Interestingly , an active ODC variant with a mutant ODC Az binding site is stable under basal degradation conditions . ^^^ |

We describe the cloning of a mouse AZI cDNA , encoding a protein with high homology to mouse ODC . ^^^ Using purified recombinant proteins , we show that AZI ( which has no ODC activity ) can release enzymically active ODC from antizyme suppression in vitro . ^^^ We also show that ODC reactivation takes place in mouse fibroblasts upon transient transfection with an AZI expressing plasmid construct . ^^^ Finally we demonstrate that the AZI mRNA content of mouse fibroblasts increases significantly within an hour of growth stimulation , i . e . much earlier than ODC transcripts . ^^^ Our results indicate that induction of AZI synthesis may represent a means of rescuing ODC molecules that have been inactivated and tagged for degradation by antizyme , when culture conditions improve and polyamine production is needed for cell growth and proliferation . . ^^^ |
