| The Nedd 8 conjugated ROC 1 CUL1 core ubiquitin ligase utilizes Nedd 8 charged surface residues for efficient polyubiquitin chain assembly catalyzed by Cdc 34 . ^^^ Here we report that the conjugation of Nedd 8 to ROC 1 CUL1 , a subcomplex of the SCF ROC 1 E3 ubiquitin ligase , selectively stimulates Cdc 34 catalyzed lysine 48 linked multiubiquitin chain assembly . ^^^ We have further demonstrated that separate regions within the human Cdc 34 C terminal tail are responsible for multiubiquitin chain assembly and for physical interactions with the Nedd 8 conjugated ROC 1 CUL1 to assemble extensive ubiquitin polymers . ^^^ These results suggest that Nedd 8 charged surface residues mediate the activation of ROC 1 CUL1 to specifically support Cdc 34 catalyzed ubiquitin polymerization . . ^^^ |

| Here , we show that the activity of human Cdc 34 in the Ub Ub ligation reaction was enhanced dramatically by SCF ' s core Ub ligase module , composed of a heterodimeric complex formed by the ROC 1 RING finger protein and the CUL 1 C terminus that contains a Nedd 8 moiety covalently conjugated at K 720 . ^^^ |
